Use the agar. Dip the loop into the agar so it breaks the surface, and it'll get kinda wet/sticky. Then start your well away from the hole in the agar. (The loop will get stuck and tear the agar further and it'll get all messy) I'm gonna have to read the rest of this post, to figure out what you are actually trying accomplish. Pick a bit of mycelia, and start that on a new clean plate. Um, one thing that you may have to look out for. The strain may get 'used to' your agar/ingredients and find it difficult to jump off onto another substrate. If you put a small bit of the final target substrate into you purity plates, that may help. Also If you keep on subbing onto new plates, your original culture may not be as vigourous as you hope. (Senescence (sp) ) I think this is called. i.e getting old. What is it your actually trying to do? What have you got and what do you want? Kai. Actually, what you can also do is: A bit difficult to describe, but useful if you're in a hurry or using a limited amount of disposable loops. With one loop, make a well of spores/bacteria. i.e a just spread bunch around a bit, keeping the loop at the same angle. Then with the loop at the same angle, smeer, say 5 lines across the agar, then, turn the loop over(so you've got a clean side, smeer 3-4 lines across the first 5 lines. Then wiggle a line using the edge of the loop, (i.e the smallest surface are of the loop), acroos the 3 latest lines. Hope that makes sense, Kai. So less farting about with disinfecting etc.. Practice, and you'll get better, and develop your own technique.

Yeah, plastic loops are generally disposable. But as Shroomy said, you could just resterlise them in 70% EtOH. k.

Hi, Fortunatly some of us work in Labs ;-) Agar, can be found in your local Chinese grocer type thing, just ask, sometimes called Agar-Agar. (On a side note, anyone try gelatin. Agar is basically non-nutricious, just a setting agent. The gelstrenght is maybe something to play around with. Actually, come to think of it, we use Lactose Gelatin, for Clostridia at work. So I'd gues it could work instaed of agar, anyway.. err, potatoes, Marck, good quality German tatties. Get 50Kgs sent from Germany, so we all can share. Actaully, there maybe a local supplier. Petri dishes, got some from Crown Sci. Plus plenty around the Web. I do believe Reville used to supply ready poured plates. Loops, Um, maybe u could make something, doesn't really have to be a loop, just something (flame) sterilisable, a good spreader, and smoothish, so it doesn't tear the agar. No need for Platinum loops. k.

They use mycelium, for bio/mycoremediation. They must be pretty effecient at transfering some things throughout their network and possible into their caps, eg the heavy metals left over from industrial land I doubt they'd transport actual pathogens, however their by-products/toxin may be tranfered. I've heard/read somewhere they use the mycelium to destroy pathogens in slurry near weatercourses. I alos doubt they'd eat the resulting mushrooms. Aspergillus is not necessarily harmless. e.g. A. niger k. Anyway, why risk it. Alcohol is quite effective at disinfection, however any toxins, would maybe remain. [ 25. Nice pretty flower there. Torsten, how many seeds do you have? Any idea how quick it grows. I cant picture how a pharm. co. could have a crop of column cacti. Would look quite weird. k.

How can u be aware of the fun potential of mushrooms without being aware of the fatal potential of mushrooms? I always thought fungipobia was an inate attribute that eventually wears off. Did anyone like (normal) mushrooms when they were kids? k.

By the way, anyone know of a relatively simple key for all mushrooms. Just something to help start an identification, then you can go on to further investigations. I've recently been taking pix of any cool mushrooms I find and want to try to start to identify some, but its difficult to start, without some kind of reference.
