  1. Different people have different iodeas of what is interesting, Torsten. There are plenty of chemicals I like the look of that I would never bother anyone about unless they were as obsessive as I am.

Thanks, friendly. I thought he was only dealing with the very simple isoquinolines. With the lotuses and poppies involved things become far more intriguing.

  2. The significance of the article abstracted below is that buprenorphine is an inhibitor of the other (than MAO) major drug-metabolising enzyme, cytochrome P450. This enzyme, found in the liver, metabolises over a third of all pharmaceuticals. It is almost certainly involved in the metabolism of DMT. The figures appear to suggest that 6 mg/l is sufficient to cause the effect so I am not sure why they should say it is insignificant.

When had she taken her last dose of buprenorphine? Considering the medication her general health and liver status may be compromised?

    Umehara K, Shimokawa Y, Miyamoto G.

Biol Pharm Bull. 2002 May;25(5):682-5. "Inhibition of human drug metabolizing cytochrome P450 by buprenorphine."

Department of Drug Metabolism, Drug Safety Research Center, Tokushima Research Institute, Osaka Pharmaceutical Co, Ltd, Kagasuno, Japan. [email protected]

The effects of buprenorphine, a powerful mixed agonist/antagonist analgesic, on several cytochrome P450 (CYP) isoform specific reactions in human liver microsomes were investigated to predict drug interaction of buprenorphine in vivo from in vitro data. The following eight CYP-catalytic reactions were used in this study: CYPlA1/2-mediated 7-ethoxyresorufin O-deethylation, CYP2A6-mediated coumarin 7-hydroxylation, CYP2B6-mediated 7-benzyloxyresorufin O-debenzylation, CYP2C8/9-mediated tolbutamide methylhydroxylation, CYP2C19-mediated S-mephenytoin 4-hydroxylation, CYP2D6-mediated bufuralol 1'-hydroxylation, CYP2E1-mediated chlorzoxazone 6-hydroxylation, and CYP3A4-mediated testosterone 6beta-hydroxylation. Buprenorphine strongly inhibited the CYP3A4- and CYP2D6-catalyzed reactions with Ki values of 14.7 microM and 21.4 microM, respectively. The analgesic also weakly inhibited specific reactions catalyzed by CYP1A1/2 (Ki=132 microM), CYP2B6 (Ki=133 microM), CYP2C19 (Ki=146 microM), CYP2C8/9 (IC50>300 microM), and CYP2E1 (IC50>300 microM), but not CYP2A6 mediated pathway. In consideration of the Ki values obtained in this study and the therapeutic concentration of buprenorphine in human plasma, buprenorphine would not be predicted to cause clinically significant interactions with other CYP-metabolized drugs.

  3. Oh dear, what a surprise.

    http://news.bbc.co.uk/2/hi/health/3527779.stm

For more depth, the original press releases:

    http://www.incb.org/e/press/2004/index.htm

Some might be particularly interested in the concept of "controlled delivery":

“Controlled delivery” means the technique of allowing illicit or suspect consignments of narcotic drugs, psychotropic substances or precursors, to pass out of, through or into the territory of one or more countries, with the knowledge and under the supervision of their authorities, with a view to identifying people involved in committing offences.

The smiley at the top is because I am having a nice day, hope you all are too.

  4. No place more apt for the discussion, old Fruit.

For the information on chemicals in certain plants the best sources online are probably shops like SAB and Maya, as well as the non-profit Erowid.

Strong alcohol usually answers for most of the extractions that are interesting. Treat with ammonia first in the case of alkaloids like those in Sinicuichi and kratom. Try not to blow yourself up.

See Auxin's page on Sinicuichi alkaloids http://www.shaman-australis.com/~auxin/heimia.html

    Keep on asking questions.

  5. I get more upset by the "my entheogen-taking is more spiritual and worthy than your drug-taking" attitude.

Taking the drink slowly over an hour or more is the advice I was given. Remember peyote chewers take their dose in small pieces over the night for the regular rituals.

The first shall be as the last.

  6. Bit pugnacious for a young student aren't you, Cactus Master? Perhaps a little more maturity from you?

Stating your "opinion" about the motivations of others whom you have never met and know nothing about might seem valid to you. To me it seems silly and adolescent. The Lycaeum welcomes that type of attitude but hopefully here things are a little more sympathetic.

  7. Conventional and wisdom seldom go together.

UV light is quite specific for turning CBD to THC, presumably a specific wavelength within the sunbed spectrum. White light is a different creature entirely.

    a) would be unlikely in alcohol. Better in solid state.

    B) is a cyclisation with dehydration, of course.

    c) Surely a positional rather than optical isomerisation? The double bond position changes.

    d) better called a dehydrogenation?

Why in alcohol? Butane is a better solvent except for its ecological effects. Ethyl acetate would be good for acetylating.

  8. I know several people who admit they were hiding behind psychedelics and it fucked them up long term.

Perhaps it indicates people comfortable with their mind but not with other people?

Depends on what you mean by multiple use. I also knew someone who spent some time in an asylum believing he was a werewolf.

  9. If anyone is stupid enough to take research chemicals without researching them after reading IN THIS ARTICLE that some people have died perhaps we don't need such people in the meme and gene pools. They were warned, let them darwin.

Why does this article not count as research for people who are interested when you think that anything put on the net is valid? Many net articles are stupid, confused and at times malicious. This is at least well-informed and balanced.

If you are really discapacitated mentally by all means spend your life in a padded, assisted institution. I want to live in a world that lives.

My only quibble is that he did not mention PMA, the perfect example of a designer drug that should never have been made available commercially. And killed quite a few people.

  10. Probably a doctor would accept that you have tried St John's Wort and it made your skin peel and itch as a better reason than possible MAOI interactions that are seldom a problem with Hypericum unless you are taking powerful drugs.
