Isotoma petraea tissue culture

i recently got into fiddling with tissue culture, heres some Isotoma plants propagated via TC from seed collected near Alice Springs. pretty happy so far!

a very simple "protocol" was used, in fact the plants shown above were grown in 1/2 MS media from all stages from seed germination to deflasking. I am doing some experiments with BAP to see if I can increase shoot multiplication.

Nice, I remember finding them growing out there once and started growing them from seed too. Such a cute native plant.

I never knew you could just sow seeds in agar like that :)

Hell yes, one of the easiest ways to start a culture off as seed coats are much more resistant to the bleach solution frequently used in plant TC for surface sterilisation.

Complex plant tissues can be much more challenging and often contain a higher rate of surface contaminants because of increased complex surface areas

One of the best/ easiest ways to start to learn plant-TC is to start with Acacia seed, I've mentioned it in several previous threads here

Here's first plant after a month or so hardening off, tis setting up a few reproductive organs already :lol: . deflasked a few from the variegated line but they all crapped out in a few days. Any tips on deflasking variegated plants DL? i imagine they may require some nutritional support.. will get around to a lit search at some stage, TC project been on backburner for a couple weeks now due to less interesting committments :BANGHEAD2:

few more on the way. The empty cells were variegated, and up the back are a few L. tupa.

So cool, thanks for the follow up :) Am happy for you that you got them to grow out and full cycle!

I have never deflasked variegated plants, I can't imagine what the problem might be- let us know if you find a workaround
