ferret
a very simple "protocol" was used, in fact the plants shown above were grown in 1/2 MS media from all stages from seed germination to deflasking. I am doing some experiments with BAP to see if I can increase shoot multiplication.

Halcyon Daze
Nice, I remember finding them growing out there once and started growing them from seed too. Such a cute native plant. I never knew you could just sow seeds in agar like that <___base_url___>/uploads/emoticons/default_smile.png
Darklight
Ferret those are *lovely*, I bet you are proud. How are they going?I never knew you could just sow seeds in agar like that https://www.shaman-australis.com/forum/applications/core/interface/imageproxy/imageproxy.php?img=&key=ed93ee4b8a158835e0af19ead9c794b11af03de360911f7858b9da338588c45dHell yes, one of the easiest ways to start a culture off as seed coats are much more resistant to the bleach solution frequently used in plant TC for surface sterilisation. Complex plant tissues can be much more challenging and often contain a higher rate of surface contaminants because of increased complex surface areasOne of the best/ easiest ways to start to learn plant-TC is to start with Acacia seed, I've mentioned it in several previous threads here Edited August 8, 2013 by Darklight
ferret
They're hardening off well I thinkOne seedling turned out to be variegatedalso similar results so far with Lobelia tupaim getting some hormone stock solutions this week, will be fun to start tweaking things a bit

Darklight
Nice! I've been thinking of trying my hand at tissue culture on some Acacia maidenii seeds I have lying about. Any recommendations Darklight?Have a look around the forums, I've posted extensively on TC. Let me know any missing info pertinent to your sitch here on this thread or a new one, Sorry, been away and a bit time pressed now, but the basics are all out thereFerret how's it going? Any progress? Cool project https://www.shaman-australis.com/forum/applications/core/interface/imageproxy/imageproxy.php?img=&key=ed93ee4b8a158835e0af19ead9c794b11af03de360911f7858b9da338588c45d
ferret
I think it's going well for my first project! Here's first plant after a month or so hardening off, tis setting up a few reproductive organs already . deflasked a few from the variegated line but they all crapped out in a few days. Any tips on deflasking variegated plants DL? i imagine they may require some nutritional support.. will get around to a lit search at some stage, TC project been on backburner for a couple weeks now due to less interesting committments few more on the way. The empty cells were variegated, and up the back are a few L. tupa. Edited September 20, 2013 by ferret
